Replay is built upon a suite of 4 main technologies: synHSV, uCell, DropSynth, and LASR. The company merges each one to develop various, potentially curative drug candidates spanning eye diseases to the brain, skin, and muscle.
synHSV allows the delivery of 30x larger payloads than AAV. However, the large size of HSVs have made them historically difficult to engineer versus AAVs or lentiviruses and there are concerns of HSVs potential to lytically replicate in the brain. So Replay is bringing new synbio tools to bring HSVs to the standards of AAV.
Then uCell enables the generation of hypoimmune cells similar to Sana. By using iPSCs, Replay can develop potentially more accessible cell therapies. And DropSynth helps create large libraries of DNA to deliver, insert into their cell therapies, and screen for protein design.
Lastly, LASR = Learned Ancestral Sequence Recognition. This is Replay’s ML toolkit to analyze the data coming out of DropSynth to search sequence space efficiently & ID therapeutic proteins and gene therapy constructs (similar to Encoded’s model for gene therapy promoters).
With these 4 tools, Replay is developing a pipeline of drugs across their product companies like Eudora spanning four main areas: eye, brain, skin, and muscle. For the eye, Replay is working on retinitis pigmentosa, a progressive disease that leads to rod photoreceptor dysfunction and ultimately vision loss. Luxturna from Spark Therapeutics was approved in 2017 for a related disease (RPE65 retinal dystrophy) but a lot more work needs to be done for patients. And in skin, Krystal Biotech announced positive data from a pivotal trial in 2021 for their HSV candidates in a skin disease, dystrophic epidermolysis bullosa. Given its business model, Replay can create a customized target-product profile for each of their programs.
