Morphic Therapeutic is a clinical-stage company developing small molecules to replace the multi-$B market of integrin targeting biologics. Founded in 2014 by Timothy Springer (discovered integrins), Morphic is taking on the daunting task of drugging specific states with oral drugs.

The MInT platform is powered by Schrödinger to map out 100s of structural conformations for the 24 integrins. The key problem is the affinity spread (>700x) between inactive & active states. This leads to design of high affinity binders that might lead to on-target safety issues.
Morphic has a phase 2 asset focused on α4β7 for IBD. Partnerships with AbbVie/Janssen have ended due to seemingly tox issues, but Morphic is the best positioned to solve these problems and keep the integrin revival going since Entyvio’s approval in 2014 for UC/Crohn's patients.
